I'm building a square box garden on my rooftop patio. I've got one box 4x4x1. Currently my patio is covered in cement pavers. Do I need to put anything down beneath the box? I've already put a 1/4 inch pressed board layer that I've drilled holes into at the base of the box. Should I be putting plastic beneath the box as well.

Welcome! I don't know if I can answer your question because I've never tried anything like that, but since your box is on cement pavers, it certainly won't hurt the pavers. The bottom of the box would eventually decompose, but if you have the box where you want it, that shouldn't be a problem. Good luck and show us a picture when you get it planted. This looks like fun!

You might consider lining the box with a heavy plastic - this will help slow decomposition of the bottom and also retain moisture - I am thinking that your rooftop will get quite warm??

I would probably put some pavers under the box at the corners; so the box is and inch or two above the pavers on the ground. This would help it drain better. Just an idea.Even a couple of boards under the box would put it up a few inches, cheaper too.
